چکیده

Immunotherapy for glioblastoma (GBM) is the subject of numerous clinical trials, given the potential for the adaptive immune response to combat this diffusely infiltrating tumor. However, rational application of immunotherapy to these tumors is challenging because of the peculiar immune privilege of the brain and the molecular heterogeneity of glioma antigens. Little is known about patient-to-patient variability in the potential to generate anti-glioma immune responses of their systemic and local T cell populations. We conducted a foundational study of the populationwide characteristics of both the peripheral blood and local, tissue-infiltrating T lymphocytes (TILs) of glioma patients using TCRseq. Through reverse transcription and pan-repertoire amplification of the TCR-alpha and -beta chains we generated TCRseq libraries from matched surgical tissue and peripheral blood cells. Using peripheral blood samples from primary GBM, low-grade glioma, and non-tumor patients (N=14), we define new immunophenotypes using statistical metrics based on

RTCR: a pipeline for complete and accurate recovery of T cell repertoires from high throughput sequencing data

MOTIVATION High Throughput Sequencing (HTS) has enabled researchers to probe the human T cell receptor (TCR) repertoire, which consists of many rare sequences. Distinguishing between true but rare TCR sequences and variants generated by polymerase chain reaction (PCR) and sequencing errors remains a formidable challenge. The conventional approach to handle errors is to remove low quality reads,...

مروری برتکنیک های توالی یابی D‏NA (نسل اول، نسل دوم و نسل سوم)

Introduction: The DNA sequencing is the most important technique in molecular biology by which the order of the nucleotides can be identified in a piece of DNA. There are several different methods for sequencing the DNA. Now, the DNA sequencing has great importance in the medical diagnostics and other medical fields. Some methods have been invented to speed up and increase the efficiency of the...

Use of high throughput T cell receptor (TCR) DNA sequencing to characterize T cell infusion products and track clonal expansion in vivo

Background During the process of T cell priming to peptide antigens, the T cell receptor (TCR) undergoes gene rearrangements that result in a peptide-binding region (complementarity determining region, CDR) which can act as a unique molecular fingerprint for similar T cell clonotypes. The purpose of this study was to assess high-throughput TCR DNA sequencing to analyze qualitative and quantitat...

Basic research and clinical application of immune repertoire sequencing

The adaptive immune system uses several strategies to generate a repertoire of Tand B-cell antigen receptors (TCR/BCR) with sufficient diversity to recognize the wide array of potential pathogens. The Immunoglobulin and TCR repertoires are a mirror of the human immune system, which reflects processes caused by infections, cancer, autoimmunity and aging. The rapid advances in sequencing technolo...

Tumor-associated T cell receptor repertoires in low- and high-grade gliomas

Glioblastoma (GBM) remains prognostically dismal, with care centered on resection, motivating research into novel therapies. Although inducing anti-tumor immunity remains an attractive target for therapeutic and preventative intervention, the interplay between evolving dysregulation of the glioma microenvironment and T cell inefficacy remains poorly understood. In our murine model of proneural ...
